His lab investigates genetic, biochemical and structural aspects of pathways involved in the maintenance of genomic stability, as well as the mechanistic basis for spontaneous and DNA damage-induced nucleotide substitution mutagenesis. The knowledge gained is used to study the involvement of these pathways in fitness and disease, including cancer, using mouse models. This work provides the basis to improve prevention and treatment of human disease.

He was amongst the first to identify mammalian mismatch repair genes and the first to generate mouse models to study the phenotypic consequences of mismatch repair gene defects for cancer predisposition. A major line of research in his laboratory is the development and validation of in vitro and in vivo assays to classify human variants in the human mismatch repair genes.
